We describe the synthesis of a polycatenated cyclic polymer, a structure that resembles a molecular charm bracelet. Ruthenium-catalyzed ring-opening metathesis polymerization of an aminocontaining cyclic olefin monomer in the presence of a chain transfer agent generated an alpha,omega-diazide functionalized polyamine. Cyclization of the resulting linear polyamine using pseudo-high-dilution coppercatalyzed click cyclization produced a cyclic polymer in 19% yield. The click reaction was then further employed to remove linear contaminants from the cyclic polymer using azide- and alkyne-functionalized scavenging resins, and the purified cyclic polymer product was characterized by gel permeation chromatography, 1H NMR spectroscopy, and IR spectroscopy. Polymer hydrogenation and conversion to the corresponding polyammonium species enabled coordination and interlocking of diolefin polyether fragments around the cyclic polymer backbone using ruthenium-catalyzed ring-closing olefin metathesis to afford a molecular charm bracelet structure. This charm bracelet complex was characterized by 1H NMR spectroscopy, and the catenated nature of the small rings was confirmed using two-dimensional diffusionordered NMR spectroscopy.

The kinetics of intermolecular ene-yne metathesis (EYM) with the Hoveyda precatalyst (Ru1) has been studied. For 1-hexene metathesis with 2-benzoyloxy-3-butyne, the experimental rate law was determined to be first-order in 1-hexene (0.3-4 M), first-order in initial catalyst concentration, and zero-order for the terminal alkyne. At low catalyst concentrations (0.1 mM), the rate of precatalyst initiation was observed by UV-vis and the alkyne disappearance was observed by in situ FT-IR. Comparison of the rate of precatalyst initiation and the rate of EYM shows that a low, steady-state concentration of active catalyst is rapidly produced. Application of steady-state conditions to the carbene intermediates provided a rate treatment that fit the experimental rate law. Starting from a ruthenium alkylidene complex, competition between 2-isopropoxystyrene and 1-hexene gave a mixture of 2-isopropoxyarylidene and pentylidene species, which were trappable by the Buchner reaction. By varying the relative concentration of these alkenes, 2-isopropoxystyrene was found to be 80 times more effective than 1-hexene in production of their respective Ru complexes. Buchner-trapping of the initiation of Ru1 with excess 1-hexene after 50% loss of Ru1 gave 99% of the Buchner-trapping product derived from precatalyst Ru1. For the initiation process, this shows that there is an alkene-dependent loss of precatalyst Ru1, but this does not directly produce the active catalyst. A faster initiating precatalyst for alkene metathesis gave similar rates of EYM. Buchner-trapping of ene-yne metathesis failed to deliver any products derived from Buchner insertion, consistent with rapid decomposition of carbene intermediates under ene-yne conditions. An internal alkyne, 1,4-diacetoxy-2-butyne, was found to obey a different rate law. Finally, the second-order rate constant for ene-yne metathesis was compared to that previously determined by the Grubbs second-generation carbene complex: Ru1 was found to promote ene-yne metathesis 62 times faster at the same initial precatalyst concentration.

Regioselectivity in the intramolecular Heck reaction of a series of N-sulfonyl-2,5-dihydro-3-substituted pyrroles was studied. These substrates are unbiased in terms of the formed ring size of the new heterocycle. Results indicate that high levels of regioselectivity are observed under a range of conditions, and that there is an underlying propensity for carbon-carbon bond formation at the most hindered end of the alkene. Conversion of essential-oil allylbenzenes (phenylpropenoids) to high-value fine chemicals via isomerization-metathesis is reported. The target reaction sequence involves isomerization of ArCH2CH=CH21 into the corresponding conjugated olefins 2, and ensuing cross-metathesis with acrylates to generate ArCH=CHCO2R 3. The second-generation Hoveyda catalyst HII was chosen for the metathesis step. A range of lead candidates was assessed for the isomerization step, of which most active was the Grotjahn catalyst [CpRu(PN)(MeCN)]PF6([4]PF6; PN = 2-PiPr2-4-tBu-1-Me-imidazole). The following order of isomerization activity was determined, using the isomerization of estragole 1a to anethole 2a (Ar = p-MeOC6H4) as a probe reaction: [CpRu(PN)(MeCN)]PF6> RuHCl(CO)(PPh3)3> Ru(Me-allyl)2(COD) > Pd2Br2(PtBu3)2> RuHCl(PPh3)3> RuCl3(mu2-C)(mu2,kappa1-C,eta6-Mes-H2IMes)Ru(H)(H2IMes) (the “Grubbs hydride”) > RuHCl(CO)(H2IMes)(PCy3) > RuHCl(CO)(IMes)(PCy3) > RuHCl(CO)(PCy3)2. To maximize process efficiency, a systematic comparison of orthogonal tandem catalysis versus sequential catalyst addition was undertaken, using catalysts [4]PF6and HII. The impact of each process type on product selectivity and catalyst compatibility was assessed. Selectivity was undermined in tandem isomerization-metathesis by competing metathesis of 1. Sequential catalyst addition eliminated this problem. The isomerization catalyst [4]PF6adversely affected metathesis yields when equimolar with HII, an effect traced to the imidazole functionality in [4]PF6. However, at the low catalyst loadings required for efficient isomerization (0.1 mol% [4]PF6), negligible impact on metathesis yields was evident. The target cinnamates and ferrulates were obtained in quantitative yields by coupling these steps in a one-pot isomerization-metathesis protocol.

Studies towards the total synthesis of the natural product radicicol are described that employ a late-stage esterification and aromatization by trapping a ketene intermediate. The subsequent biomimetic aromatization of the resultant triketo ester gave highly functionalized resorcylates. Two distinct methods were examined that trap the ketene intermediate through either an intermolecular or intramolecular process. In the first approach, the synthesis of the resorcylate was followed by a ring-closing metathesis, which gave the macrolactone and protected precursors to monocillin I. In the second approach, an intramolecular ketene trapping was examined as an alternative to close the macrocycle and form the resorcylate macrolactone.
